Лаборатория физиологической генетики ИБГ УФИЦ РАН / Laboratory of Physiological Genetics IBG UFRC RAS
Лаборатория организована в 2000 г. как лаборатория биохимической генетики человека. В связи с расширением области исследований в 2008 г. лаборатория переименована в лабораторию физиологической генетики. В 2011 г. в состав лаборатории вошла лаборатория экологической генетики человека (зав. лаб д.м.н., проф. Викторова Т.В.).
С 2019 года руководителем лаборатории является доктор биологических наук, доцент Корытина Гульназ Фаритовна.
The laboratory was created in 2000 and initially was focused on human biochemical genetics. Expansion of the research scope led to the transformation of the laboratory in 2008 into Laboratory of Physiological Genetics. In 2011, the laboratory was joined with the Laboratory of Human Ecological Genetics (head of the laboratory – Prof. Viktorova TV, MD).
Since 2019, the head of the laboratory has been Associate Professor Korytina Gulnaz Faritovna, PhD.

Основные направления научной деятельности / The main directions of scientific activity
В лаборатории физиологической генетики в течение двадцати лет проводятся исследования фундаментальных основ сложно-наследуемых признаков, приводящих к ограничению продолжительности жизни, и молекулярно-генетических факторов долголетия. Осуществляются систематические исследования молекулярно-генетических основ многофакторных заболеваний сердечно-сосудистой, нервной, эндокринной, дыхательной систем. Исследование полиморфизма и функциональной активности генов на выборках клинически здоровых и больных людей разного возраста, включая долгожителей, важны для поиска триггеров старения, раскрытия тонких молекулярных механизмов потенциально патогенетических изменений, развивающихся в организме по мере старения и влияющих на продолжительность жизни, риск развития, возраст манифестации и прогноз исхода широкого спектра заболеваний разных систем организма. Результаты научных работ лаборатории также имеют большое значение для разработки системы маркеров биологического возраста, поиска потенциальных мишеней для геропротекторов. В лаборатории собраны обширные коллекции биологического материала человека и модельных организмов (комнатной мухи и колорадского жука).
Лаборатория физиологической генетики располагает архивом анкетных данных и коллекцией образцов биологического материала (кровь, ДНК, кДНК, сыворотка) жителей различных этнических групп Республики Бащкортостан – башкир, русских, татар. База сложно-наследуемых заболеваний включает образцы ДНК лиц с верифицированными диагнозами, такими как хроническая сердечная недостаточность, инфаркт миокарда, гипертоническая болезнь, ишемический инсульт, сахарный диабет 2 типа, сахарный диабет 1 типа, рассеянный склероз, острая коронарная недостаточность, хроническая обструктивная болезнь легких, профессиональный хронический бронхит, метаболический синдром, ожирение, нарушения пищевого поведения. Контрольная выборка здоровых индивидов включает группы новорожденных, лиц среднего, пожилого, старческого возраста, а также долгожителей до 110 лет. В целом коллекция лаборатории насчитывает свыше 6000 тысяч образцов ДНК.
For over twenty years, the Laboratory of Physiological Genetics has been focusing on the study of molecular mechanisms of complex traits, including aging-associated diseases limiting the lifespan, and genetic factors of longevity. Areas of research include molecular pathways of complex diseases of the cardiovascular, nervous, endocrine, and respiratory systems. The study of polymorphism and functional activity of genes in the cohorts of healthy people and patients with various diseases of different ages, including long-livers, is important for the identification of triggers of aging, unveiling of subtle molecular mechanisms of potentially pathogenic aging-associated changes that affect life expectancy, risk of development, age of manifestation and prognosis of the outcome of a wide range of diseases. The results of the research being conducted in the laboratory are also of great importance for the detection of markers of biological age, the search for potential targets for geroprotectors. The laboratory has collected extensive collections of human biological material and model organisms (housefly and Colorado potato beetle).
The Laboratory of Physiological Genetics has an archive of questionnaire data and a collection of samples of biological material (blood, DNA, cDNA, serum) inhabitants of various ethnic groups of the Republic of Baschkortostan – Bashkir, Russians, Tatars. The datebase of complex diseases includes DNA samples of persons with verified diagnoses, such as chronic heart failure, myocardial infarction, arterial hypertension, ischemic stroke, type 2 diabetes mellitus, type 1 diabetes mellitus, multiple sclerosis, acute coronary insufficiency, chronic obstructive pulmonary disease, occupational chronic bronchitis, metabolic syndrome, obesity, eating addiction disorders in adults and children. The control sample of healthy individuals includes groups of newborns, collection of the laboratory has over 6,000 thousand DNA samples.
Важнейшие научные результаты / The most important scientific results.
Исследованы особенности генетической архитектуры ассоциированных с возрастом заболеваний сердечно-сосудистой, нейроэндокринной и дыхательной систем с использованием в качестве маркеров полиморфных участков более 200 генов-кандидатов. Идентифицированы геномные маркеры предрасположенности к ассоциированным с возрастом заболеваниям, выявлены гены, имеющие потенциальную значимость как маркеры старения и мишени для геропротекторов. Выявлены паттерны генов, дифференциально экспрессирующихся в мононуклеарных клетках крови людей старческого возраста и долгожителей, пациентов с хронической обструктивной болезнью легких, сердечно-сосудистыми заболеваниями. Подтверждена гипотеза о роли полиморфизма, обусловленного инсерцией или делецией Alu-ретроэлементов в интроны генов, в вариабельности продолжительности жизни человека. Сотрудники лаборатории принимают активное участие в работе отечественных и международных конференций в форме устных докладов и стендовых сообщений, читают курс лекций по современным проблемам цитогенетики, медицинской генетики, экологической генетики для студентов Башкирского Государственного Медицинского Университета и Башкирского Государственного Педагогического Университета, являются научными руководителями студенческих научных кружков, курсовых и дипломных работ, выполняемых на базе лаборатории. Исследования проводятся в рамках выполнения Программ фундаментальных научных исследований государственных академий наук, а также проектов, поддержанных Российским Фондом Фундаментальных Исследований, Российским Гуманитарным Научным Фондом, Академией Наук Республики Башкортостан, Федеральной Целевой Программой, Министерством науки и высшего образования РФ.
The members of the laboratory have defined the genetic architecture of age-associated diseases of the cardiovascular, neuroendocrine and respiratory systems using more than 200 polymorphic markers of candidate genes. Genomic markers of predisposition to age-associated diseases have been detected, and genes that have potential significance as markers of aging and targets for geroprotectors have been identified. The patterns of genes that are differentially expressed in blood mononuclear cells of people of old age and long-livers, patients with chronic obstructive pulmonary disease, cardiovascular diseases have been revealed. The hypothesis on the role of polymorphism caused by insertion or deletion of Alu retroelements into gene introns in the variability of human life span has been confirmed. The members of the laboratory actively present their findings at Russian and international scientific meetings, read courses of lectures on modern problems of cytogenetics, medical genetics, ecological genetics for students of the Bashkir State Medical University and the Bashkir State Pedagogical University, are the scientific leaders of student scientific societies, supervise Bachelor, Master, and PhD students. Research is performed within the framework of the programs of fundamental scientific research of the state academies of sciences, as well as projects supported by the Russian Foundation for Basic Research, the Russian Humanitarian Scientific Foundation, the Academy of Sciences of the Republic of Bashkortostan, the Federal Target Program, the Ministry of Science and Higher Education of the Russian Federation

Research interests are related to the study of the genetics of complex diseases, in particular, chronic respiratory diseases (chronic obstructive pulmonary disease, childhood lung diseases, occupation asthma and chronic bronchitis), metabolic disorders (type 2 diabetes mellitus, obesity), cardiovascular diseases and the molecular basis of human adaptation to the conditions of chronic anthropogenic pollution, as well as molecular patterns of aging. The current collection of biomaterials of patients with chronic obstructive pulmonary disease, children with chronic respiratory diseases, workers with professional diseases of respiratory and healthy individuals contains more than 2000 DNA samples and 200 RNA samples belonging to the main ethnic groups living in the Republic of Bashkortostan. At present, our laboratory work on the study of the patterns of formation of a pathological phenotype in connection with expression, gene polymorphism, and epigenetic processes. Has been working in IBG since 2001, main scientific interests are focused on the study of the genetics of multifactorial diseases, in particular occupational and work-related diseases in workers. Currently, O.V. Kochetova is researching the genetic basis of type 2 diabetes, obesity and metabolic syndrome, eating addiction disorders in adults and children. С 2017 года является доцентом и заведующей учебной частью кафедры медицинской генетики и фундаментальной медицины Башкирского государственного медицинского университета. Эрдман Вера Викторовна, к.б.н., с.н.с., начала работать в ИБГ с 2002 года, ведет активную научную работу в области генетики долголетия и старения человека. Научные интересы Эрдман В.В. связаны, главным образом, с изучением роли структурных и функциональных особенностей генов клеточных процессов в формировании статуса долгожительства и в развитии многофакторных социально-значимых заболеваний и старения. С 2003 года Эрдман В.В. проводит популяционно-генетическое исследование долголетия в Республике Башкортостан. Актуальная коллекция содержит более 3000 образцов ДНК и РНК, собранных с привлечением неродственных между собой мужчин и женщин в возрасте от рождения до 113 лет, принадлежащих к основным этническим группам, проживающим на территории Республики Башкортостан. Эрдман В.В. имеет 87 научных работ, посвященных, в частности, исследованию влияния генетической нестабильности на продолжительность жизни поиску ассоциаций с долголетием полиморфных SNP-локусов ряда генов транскрипционных факторов и белков, вовлеченных в процессы фармакологического ответа, антиоксидантной защиты, липидного и воспалительного ответа. В настоящее время проводится поиск ключевых генов и генных сетей, участвующих в формировании этноспецифического и универсального стабильного высокоадаптивного фенотипа долгожителя. Erdman V.V. has 87 scientific papers concerned, in particular, to the study of the influence of genetic instability on lifespan to the search of the associations of longevity with polymorphic SNP-loci of the genes encoded transcription factors and proteins involved in pharmacological response, antioxidant protection, lipid metabolism, inflammatory response, have been established.
